Isolation and identification of C. perfringens from suspected cases of enterotoxemia in Fars province.

Enterotoxemia is one of the frequent diseases of domestic livestock in Iran and other parts of the world. Clostridium perfringens is the causative agent and is classified into five types (A to E ) based on the production of 4 typing toxins ( , , and ) .These toxins have important role in pathogenesis of the disease. High amounts of these toxins along with many bacteria are present in the intestinal contents of dead or affected animals. Detection of toxin in intestinal contents constitutes definitive evidence of enterotoxemia. This study was done in purpose of isolation and identification of different toxinotypes of Clostridium perfringens responsible for enterotoxemia in Fars province by culture and Elisa methods. About 167 samples of intestinal contents of sheep and goats suspected of enterotoxemia were collected and subcultured on TSC agar and neomycin supplemented blood agar .Cl. perfringens isolates were tested and typed by PCR .Presence of three toxins (alpha , beta and epsilon) were investigated in intestinal contents by Elisa method as well. In culture method 102 strains of Cl.perfringens were isolated and confirmed by biochemical tests and PCR. Toxinotyping of these isolates by PCR method revealed type A as the most common type . The second , third and fourth prevalent toxinotypes were type D, B and C respectively.Cl. perfringens type E was not detected. Compared to culture method , in Elisa method Cl. Perfringens type D was the most prevalent toxinotype involved in the disease.Same results were obtained for type B & C in both methods.
