Изучение цитотоксического влияния синтетических пиретроидов in vitro | А study of cytotoxic effect of the synthetic pyrethroids in vitro

Russian. Изучали влияние синтетического пиретроида дельтаметрина (ДМ) на перевиваемые культуры клеток животных. ДМ растворяли в 10%-ном растворе диметилсульфоксида, использовали концентрации 0,38; 0,038; 0,0038 мг/мл. В контроле к культурам добавляли солевой раствор Хенкса. Клетки инкубировали в присутствии токсина в течение 72 ч. ДМ в дозе 0,38 мг/мл вызывал гибель 83,9% клеток перевиваемой культуры НГУК-1; 0,038 мг/мл - 50,4%; 0,0038 мг/мл - 23,5%. Для перевиваемой почечной линии культуры клеток МDВК ДМ в дозе 0,38 мг/мл вызывал гибель 71,2% клеток; 0,038 мг/мл - 45,8% клеток, 0,0038 мг/мл - 15,7%. При внесении в культуральную среду ДМ в дозе 0,038 мг/мл цитотоксический эффект проявлялся снижением индекса пролиферации в 2 раза, поэтому данную дозу можно рассматривать как LC50. Предложенная модель может быть использована для решения разнообразных задач в области токсикологии и клеточной биологии, в частности для изучения физического и биологического действия ряда токсинов, представляющих практический интерес, на клеточные культуры.

English. The study investigated the effect of a synthetic pyrethroid deltamethrin (DM) on continued animal cell cultures. DM was dissolved in 10% DMSO at the rates of 0.38, 0.038, 0.0038 mg/ml. In control the cultures were added with Hank's salt solution. The cells were incubated in the presence of the toxin for 72 h. DM at the rate of 0.38 mg/ml caused the death of 83.9% cells of the continued NGUK-1 culture; at the rate of 0.038 mg/ml – of 50.4%; and at the rate of 0.0038 mg/ml – of 23.5% cells. In case of continued renal MDBK cell culture DM at the rate of 0.38 mg/ ml caused the death of 71.2% cells, the rate of 0.038 and 0.0038 mg/ml resulted in a death of 45.8% and 15.7% of cells, respectively. The cytotoxic effect from the introduction of DM in the cell culture at the rate of 0.038 mg/ml manifested in the decrease of the proliferation index in 2 times and the dose was considered as LC50. The proposed model can be used to solve various tasks in the field of toxicology and cell biology, specifically for investigation of physical and biological effect of a number of toxins of practical interest on cell cultures.
