Интеграция световой обработки в технологию получения дигаплоидов озимой пшеницы | Integration of light treatment in production technology of dihaploids of winter wheat

English. The aim of this study was to investigate the effect of the integrated light treatment at 24 h photoperiod and 25 degrees on the induction of embryogenesis in anther culture of F1 hybrids of winter wheat and on gynogenesis at chromosome elimination method. For complex lighting we use Gro-lux and Coralstar lamps. In 30 days at complex lighting plants were transferred to 16-hour daylight. Anthers with mononuclear pollen were transferred on NPB- 99 medium with complex lighting until embryogenesis. We reseeded embryoids on a medium 190-2 and grew them at 16-hour photoperiod, and then regenerants were reseeded onto growth medium under the same conditions. Developed regenerants were treated during 6 h by solution of 0.3% colchicine. In the chromosome elimination method 2 days after castration of the flowers, they were pollinated by maize and treated with 2.4-D (100 mg/l) and placed under illumination systems. On the 16th day embryoids were isolated and transferred to 190-2 medium under complex lighting. Regenerants were planted in containers and placed for 20 days under lighting systems, then for 16-hour daylight to obtain grains. In control experiments, it was 16-hour daylight. In anther culture method with integrated light treatment we received 69 dihaploids and 22 dihaploids in the control; the number of dihaploids was 3.1 times more. By the integrated light treatment chromosome elimination method we received 43 dihaploids and 13 dihaploids in the control. During embryogenesis in anther culture with the light treatment we received 563 embryoids, in the control–352 ones. By the chromosome elimination method we obtained 128 and 120 embryoids, respectively. Light treatment increases the dihaploid yield in both methods 3 times, the maximum effect was observed in the induction of embryogenesis in the anther culture.

Russian. Целью работы было исследование действия комплексной световой обработки при 24 ч световом дне и 25 град. С на индукцию эмбриогенеза в культуре пыльников гибридов F1 озимой пшеницы и на гиногенез при методе гаплопродюсера. Для комплексного освещения брали лампы Gro-lux и Coralstar. Через 30 дн. при комплексном освещении растения переносили на 16 ч световой день. Пыльники с одноядерной пыльцой вычленяли, переносили на среду NPB-99 под комплексное освещение до появления эмбриоидов. Их пересевали на среду 190-2 и выращивали при 16 ч световом дне, затем регенеранты пересевали на ростовую среду при тех же условиях. Развитые регенеранты обрабатывали 6 ч 0,3% раствором колхицина. При использовании метода гаплопродюсера через 2 дн. после кастрации цветки опыляли кукурузой, обрабатывали 2,4-Д (100 мг/л) и помещали под комплексное освещение. На 16-й день вычленяли зародыши и переносили на среду 190-2. Регенеранты высаживали в сосуды и помещали на 20 дн. в такие же условия, затем на 16 ч световой день до получения зерновок. В контроле обоих опытов был 16 ч световой день. Методом культуры пыльников получено 69 дигаплоидов при комплексной световой обработке и 22 дигаплоида в контроле: число дигаплоидов в тестовом варианте в 3,1 раза больше. Методом гаплопродюсера получено 43 дигаплоида при комплексной световой обработке и 13 дигаплоидов в контроле. При эмбриогенезе в культуре пыльников со световой обработкой получено 563 эмбриоида, в контроле – 352. Методом гаплопродюсера получено 128 и 120 эмбриоидов соответственно. Световая обработка повышает выход дигаплоидов при обоих методах в 3 раза, максимальный эффект отмечен при индукции эмбриогенеза в культуре пыльников.
