Selection of lactose-adapted strain of Arthrobacter sulfonivorans LF-GAL - producer of beta-galactosidase and development of medium composition for its cultivation

Методом многоступенчатой адаптации бактерий Arthrobactersulfonivorans БИМ В-2242 к лактозе получен морфологически и биохимически стабильный штамм A. sulfonivorans ЛФ-ГАЛ, который по эффективности синтеза внеклеточной р-галактозидазы (22,8 ед/мл) в 1,5 раза превосходит исходный штамм (15,2 ед/мл). Разработан состав питательной среды для глубинного культивирования штамма A. sulfonivorans ЛФ-ГАЛ, обеспечивающий повышение продукции фермента до 35 ед/мл, что в 1,5 раза превышает его исходный показатель и в 2,3 раза-показатель родительского штамма. Полученные данные будут использованы для разработки на основе селектированного штамма A. sulfonivorans ЛФ-ГАЛ технологии производства внеклеточной 6-галактозидазы.

Multistage adaptation of bacteria Arthrobacter sulfonivorans BIM B-2242 to lactose resulted in selection of morphologically and biochemically stable strain of Arthrobacter sulfonivorans LF-GAL exceeding 1.5 times the parent strain in efficiency of extracellular beta-galactosidase synthesis (22.8 u/ml vs 15.2 u/ml). Optimal composition of nutrient medium has been formulated for submerged fermentation of strain of Arthrobacter sulfonivorans LF-GAL to secure rise in enzyme productivity up to 35 u/ml, which is 1.5 times higher than its initial level and 2.3 times superior to that of the parent strain. Obtained results can be used for the development of bacterial beta-galactosidase biotechnology for food industry and feed production.
