Morphology-function state of rat adipose-derived mesenchymal stem cells under the suppression of oxidative stress

Изучено влияние эмоксипина на пролиферативную активность, жизнеспособность мезенхимальных стволовых клеток (МСК) из жировой ткани крыс в условиях культуры, а также на состояние в МСК системы анти-оксидантной защиты. Показано, что при 0,0005-0,005 % концентрациях эмоксипина в ростовой среде проли-феративная активность МСК увеличивается, поддерживается высокая жизнеспособность клеток и снижается в МСК содержание пероксида водорода и супероксид-анион-радикалов. При концентрациях 0,01-0,25 % эмок-сипин оказывает токсическое действие на МСК, что проявляется в снижении пролиферативной активности, жизнеспособности клеток и возрастании внутриклеточного содержания супероксид-анион-радикалов.

Generation of reactive oxygen species (ROS), representing essential process for cell functioning, leads to the development of oxidative stress, inducing disorder of cell functional status and cell death. Sustaining of ROS on optimal physiological level, localization of the activity zones of reactive oxygen species is carried out with the help of antioxidant cell system, including low molecular antioxidants and enzymes of antioxidant activity. For the prevention of the development of oxidative stress in cells there have been used synthetic antioxidants, influencing the system of natural antioxidant substances, changing their biological activity. The influence of emoxipine on proliferative activity and viability in rat adipose-derived mesenchymal stem cells (MSCs) in culture, as well as the state in the MSCs of the antioxidant protection system has been investigated in the course of the research. It has been shown that when 0.0005-0.005% concentrations of emoxipine in growth medium, the proliferative activity of mesenchymal stem cells increase, has not influenced the cell viability and reduces intracellular content of hydroperoxide and superoxide anion radicals. At concentrations of 0.01-0.25% of emoxipine has revealed the toxic effects on MSCs, that manifested itself in reduced proliferative activity and cell viability, increases intracellular content of superoxide anion radicals.
