Effect of amphotericine B on morphological and biochemical properties of Penicillium adametzii - producer of extracellular glucose oxidase

Изучено влияние стресс-фактора амфотерицина В на морфолого-биохимические свойства промышленного продуцента глюкозооксидазы - Penicillium adametzii. Установлено, что в результате действия антибиотиков удлиняется время прорастания спор гриба и снижается их жизнеспособность. введение амфотерицина В (1.1 x 10-8 M) в питательную среду в различные фазы роста Penicillium adametzii при глубинном культивировании приводят к снижению накопления биомассы и уровня образования глюкозооксидазы. Селекционированы по устойчивости к амфотерицину В (5.4 x 10-6 - 2.0 x 10-5 M) и охарактеризованы морфологические варианты Penicillium adametzii. Повышение уровня синтеза глюкозооксидазы на 12,5-88,2% при глубинном выращивании отмечено у 4 штаммов Penicillium adametzii А: 2.19, 5.16, 5.17, 5.18. Penicillium adametzii А 5.17 отобран как перспективный продуцент глюкоззоксидазы, характеризующийся максимальными значениями уровня образования фермента и продуцирующий способности мицелия.

The article analyzes the effect of stress factor of amphotericin B on morphological and biochemical properties of industrial glucose oxidase producer - Penicillium adametzii has been analyzed. It has been found that antibiotic action extends the time of fungal spore generation and reduces their viability. supply of amphotericin B (1.1 x 10-8 M) into the nutrient medium at various growth stages of Penicillium adametzii submerged culture resulted in retarded biomass accumulation and decreased level of glucose oxidase synthesis. Morphological variants of Penicillium adametzii were screened and characterized for amphotericin B resistance (5.4 x 10-6 up to 2.0 x 10-5 M). Enhanced glucose oxidase production (by 12.5 - 88.2%) in the course of submerged fermentation was recorded in 4 Penicillium adametzii strains: 2.19, 5.16, 5.17, 5.18. Penicillium adametzii A 5.17 was chosen as the promising glucose oxidase producer distinguished by maximum values of enzyme generation and productive capacity of the mycelium.
