Amino acid profile of cartilaginous tissue of cows at the brahyspina syndrome

English. In this work was investigated an amino acid profile of tissue of the cattle of Holstein in normal and in hereditary defect of the backbone, which caused by BS-syndrome. DNA-diagnostics of a carriage of a mutation of a gene of FANCY at farm animals was carried out by the PCR-analysis with use of the special primers. Screening of an amino acid profile of the animals was carried out by reverse phase liquid chromatography with detecting in fluorescence. The comparative analysis of content of free amino acids and their metabolites in cartilaginous tissue of cows have shown reliable increase in concentration of the main biologically active groups of free amino acids more than by 15 times in comparison with genetically healthy control. It is caused by influence of mutations of the gene of FANCY on processes of a metabolism of free amino acids in cells of tissue of cattle of Holstein.

Russian. В данной работе исследовали аминокислотный профиль крупного рогатого скота в норме и при наследственном пороке позвоночника, обусловленного развитием BS-синдрома. ДНК-диагностику носительства мутации гена FANCY у сельскохозяйственных животных проводили методом ПЦР-анализа с использованием специальных праймеров. Скрининг аминокислотного профиля животных проводили методом обращенно-фазовой жидкостной хроматографии с детектированием по флуоресценции. Сравнительный анализ содержания свободных аминокислот и их метаболитов в биологическом материале при BS-синдроме показал достоверное увеличение концентраций основных биологически активных групп свободных аминокислот более чем в 15 раз по сравнению с генетически здоровым контролем, что обусловлено влиянием мутации гена на процессы метаболизма свободных аминокислот в клетках ткани крупного рогатого скота голштинской породы.
