The infection hazard of carriers of proviral bovine leukemia virus and its evaluation with regard to leukocytosis

English. The spread of the bovine leukemia virus (BLV) does a significant economic damage to dairy and beef cattle breeding. The content of viral RNA in blood samples and the proliferative activity of were compared to study a possibility of using these indices for individually testing animals by the both characteristics. The presence or absence of genome integrated provirus DNA in Black-and-White Holstein cows (n = 57, commercial herd) was evaluated and the group of infected animals was revealed when using primers as designed for BLV gag and pol genes. The relative quantity expression of provirus in individual infected cows using reverse transcriptase OCR (RT-PCR) analysis and primers to the fragment of the BLV pol gene were assessed. The count of leukocytes in blood samples of all cows, included in the analysis, was further calculated. The number of leucocytes in cows free from infection, with the exception of one cow, was less than 12*10E9 cells/l. This indicator in infected animals divided the investigated cows into two subgroups — in the first subgroup the leucocyte values did not exceed 17*10E9 cells/l, in the second subgroup the number of leukocytes was significantly higher, reaching 28*10E9 cells/l in some animals. It was important to note, that the amount (above 20*10E9 cells/l) of BLV RNA, sufficient to detect by RT-PCR, was revealed only on the second subgroup. The coincidence of the increased leucocyte amount and the BLV RNA in blood of infected animals indicates that particular these cows represent the greatest infection hazard and suggests that the combination of the two estimates (the number of leukocytes and RNA BLV) can be quite a reliable approach to the herd recovery by priority of their liquidation.

Russian. Распространение вируса бычьего лейкоза (Bovine leukemia virus, BLV) наносит существенный экономический ущерб молочному и мясному скотоводству. Проведено сопоставление содержания вирусной РНК в образцах крови и пролиферативной активности лейкоцитов, чтобы изучить возможность использования этих показателей для индивидуального тестирования животных по обеим характеристикам. С использованием разработанных праймеров к генам BLV gag и pol у черно-пестрых голштинизированных коров (n = 57) из промышленного стада определили наличие или отсутствие провирусной ДНК BLV, интегрированной в геном, и выявили группу инфицированных особей. С помощью PCR с обратной транскриптазой (RT-PCR) и праймеров к консервативному фрагменту гена pol BLV оценили относительное количество экспрессирующегося провируса у индивидуальных инфицированных животных. Дополнительно у всех коров, включенных в анализ, подсчитали число лейкоцитов в образцах крови. Инфицированные животные по этому показателю разделились на две подгруппы: у первой значения не превышали 17*10E9 кл/л, у второй — оказались существенно выше, достигая у некоторых особей 28*10E9 кл/л. У коров, свободных от инфекции (за исключением одной особи), число лейкоцитов было ниже 12*10E9 кл/л. Оказалось, что достаточное для выявления в RT-PCR количество РНК вируса BLV обнаруживается только у второй подгруппы инфицированных коров. Повышение количества лейкоцитов (свыше 20*10E9 кл/л) в сочетании с накоплением вирусной РНК в образцах крови инфицированных животных свидетельствует о том, что именно они представляют наибольшую инфекционную опасность. Совместное использование двух показателей (число лейкоцитов и количество вирусной РНК BLV) может быть достаточно надежным подходом при первоочередных мероприятиях по оздоровлению стад крупного рогатого скота.
