Enhancement of Indole-3-Acetic acid biosynthetic capacity of Paenibacillus polymyxa CNLM12 mutants by using mini-tn10 transposon

To date, many literatures have reported that the biosynthesis of phytohormone indole-3-acetic acid (1AA) by plant-growth promoting rhizobacteria (POPR) can affect to the growth of crops. In order to enhance the capabilities of producing bacterial IA A. over 3,000 mutants were generated by random insertion of transposon mini-Tn/0 into genome of parental bacterium P. polymyxa CNLM12 with help of mediating plasmid pTC333. Among transformants tested, 12 mutants showed the increase and decrease of IAA productivity comparing with die wide-type P. polymyxa CNLM12. Two mutants ouxi203, auxi-188 exhibited the most producers of IAA, which were 57% and 62% higher than wide-type. All 12 mutants were examined the presence of transposon mini-Tn10 and analyzed the sequence of genes disrupted by mini-Tn10 by reverse-PCR technique. Innovation of each of 12 mutants onto seedling, roots of pepper (Capsicum annuum L.) and maize (Zeu mays L.) revealed that the length and weight of root, shoot and total weight of plants were increased in some treatments but not linear with the increase/decrease of IAA productivity of mutants. Interestingly, the mutant auxi488 demonstrated the highest biosynthesis of IAA and the most growth promotion of maize and pepper seedlings.
