تأثیر هیدروپرایمینگ و پیری بذر بر ویژگی‌های جوانه‌زنی و بیوشیمیایی بذر لوبیا چیتی (Phaseolus vulgaris) تحت تیمار شوری | Effect of hydropriming and seed aging on seed germination and biochemical characteristics of pinto bean (Phaseolus vulgaris) seed under salt stress

This study was conducted to evaluate the effect of hydropriming and seed aging on germination and enzymatic properties of pinto bean under salinity stress as factorial based on a completely randomized design with four replications. Two groups of seeds (i.e., non-aged and aged seeds), two hydropriming treatments (i.e., hydro primed and unprimed seeds) and six salinity treatments (i.e., 0, 2, 4, 6, 8 and 10 dS/m) were the experimental factors. The results showed that the highest mean time and percentage of germination, plumule length and vigor were observed in the control (i.e., distilled water) and hydropriming treatments. Maximum root length and percentage of seedling water were obtained in the control (i.e., distilled water) and aged seed treatments. In addition, in terms of radicle dry weight, the highest amount was observed in salinity of 4 dS/m and non-aged hydro primed seeds. The highest plumule dry weight was observed in salinity of 6 dS/m and allometric index in salinity of 8 dS/m for non-aged seed and aged seed without hydropriming, respectively. An increase in the levels of salinity stress and aging the seeds increased the malondialdehyde and reduced the activity of germination, mean time and germination percentage, and seedling growth. Seed hydropriming reduced the peroxidation of the cell membrane and generally improved the speed and uniformity of germination, aged and natural seeds vigor under both salinity and optimum conditions. As a result, hydropriming can increase the tolerance of bean seeds to salinity at the germination stage and increase the germination capacity of stored seeds for cultivation. Highlights:The effect of hydropriming on recovering the power of seed germination of pinto bean has been studied under the salinity condition.An attempt has been made to determine the effect of hydropriming on increasing the tolerance of pinto bean seeds to storage and salinity stress condition.The effect of pinto bean storage on biochemical changes and enzyme activity has been studied in salinity stress condition.

Persian. این پژوهش با هدف بررسی اثر هیدروپرایمینگ و پیری بذر بر ویژگی­های جوانه­ زنی و آنزیمی بذر لوبیا چیتی تحت­ تأثیر شوری به­ صورت فاکتوریل، در قالب طرح کاملاً تصادفی با چهار تکرار در سال 1393 انجام شد. تیمارهای آزمایش شامل دو گروه بذر، طبیعی و پیر، دو تیمار شاهد و هیدروپرایمینگ و شش تیمار شوری صفر، 2، 4، 6، 8 و 10 دسی­زیمنس بر متر بود. نتایج این آزمایش نشان داد که بیشترین متوسط زمان و درصد جوانه­ زنی، طول ساقه­ چه و بنیه بذر در تیمار شاهد (آب مقطر) و هیدروپرایمینگ و همچنین، بیشترین طول ریشه­ چه و درصد آب بافت گیاهچه در تیمار شاهد (آب مقطر) و بذر پیر به­ دست آمد. همچنین از نظر وزن خشک ریشه­ چه، در شوری 4 دسی‌زیمنس بر متر و بذر طبیعی هیدروپرایمینگ بیشترین مقدار مشاهده شد. بیشترین میزان وزن خشک ساقه­ چه در شوری 6 دسی‌زیمنس بر متر و ضریب آلومتریک در شوری 8 دسی‌زیمنس بر متر به­ ترتیب برای بذر طبیعی و بذر پیر بدون هیدروپرایمینگ مشاهده گردید. با افزایش سطوح تنش شوری و پیری بذر میزان مالون دی آلدهید افزایش یافت و از میزان فعالیت آنزیم­های جوانه ­زنی، درصد جوانه­ زنی و رشد گیاهچه کاسته شد. هیدروپرایمینگ بذر موجب کاهش پراکسیداسیون غشای سلولی و به ­طور کلی سبب بهبود سرعت و یکنواختی جوانه­ زنی، بنیه بذرهای پیر و طبیعی هم در شرایط شوری و هم در شرایط مطلوب گردیده است. در نتیجه هیدروپرایمینگ باعث افزایش تحمل به شوری بذر لوبیا در مرحله جوانه­ زنی و افزایش قدرت جوانه‌زنی بذرهای انبارداری شده جهت کشت مؤثر گردید. جنبه‌های نوآوریتأثیر هیدروپرایمینگ در بازیابی قدرت جوانه­زنی بذر انبارداری شده لوبیا چیتی تحت شرایط شور.تأثیر هیدروپرایمینگ در افزایش تحمل بذر لوبیا چیتی به شرایط انبارداری و تنش شوری.تأثیر انباداری بذر لوبیا چیتی بر تغییرات بیوشیمیایی و فعالیت­های آنزیمی در شرایط تنش شوری.
