Impact of selected factors on Trichoderma sporulation and biosynthesis of hydrolases in bioreactor | Wpływ wybranych czynników na przetrwalnikowanie i biosyntezę hydrolaz w hodowlach grzybów Trichoderma w bioreaktorze

Badania skupiły się na ocenie wartości biologicznej hodowli 6 szczepów Trichoderma w płynnym podłożu ziemniaczanym komplementowanym odpadami ligninocelulozowymi, takim jak wysłodki buraczane, otręby pszenne i wytłoki jabłkowe. Spośród testowanych szczepów po ocenie poziomu aktywności hydrolitycznych (celulaz, ksylanaz, poligalakturonaz, lichenaz i laminarynaz) wybrano 2 szczepy do hodowli bioreaktorowych z przeznaczeniem do produkcji biopreparatów. Po zakończeniu hodowli biomasę odwirowano, wraz z pozostałościami materiału ligninocelulozowego. Następnie wysuszono w strumieniu ciepłego powietrza i oceniono przeżywalność zarówno bezpośrednio po suszeniu, jak i po upływie 3 miesięcy. Szczep TRS40 charakteryzował się wyższym (72%) niż TRS43 poziomem zachowania żywotności, co może dać gwarancję uzyskania aktywnego biopreparatu.

English. The subject of the presented work were preliminary studies to obtain Trichoderma biopreparation, based on liquid culture medium. Cultures of filamentous fungi can be an alternative to solid state fermentation (SSF) in obtaining spore-based biopreparations, relevant in the protection of plants against phytopathogens. The study focused on the as- -sessment of the biological value of 6 strains of Trichoderma cultured in liquid potato medium, complemented with lignocellulosic waste, such as beet, wheat bran and apple pomace. Out of the tested strains after the assessment of the hydrolytic activity level (cellulases, xylanases polygalacturonases, lichenases and laminarinases), 2 strains were selected for cultivation in bioreactors, intended for the obtaining of a biopreparation. After the cultures, biomass was centrifuged, along with the remains of the ligninocellulosic material, dried in a stream of hot air. Survival rate of spores was assessed, both, directly after drying and after 3 months of storage. The strain TRS40 was characterized by higher viability (72%) as compared with the TRS43 strain, which ensures activity of the biopreparation.
