Oyster mushroom is mostly used edible mushroom worldwide due to dietary essential. It is exceptional among others. The distinction of substrate is significant reason for the production of mushrooms. Substrates play a meaningful role in mushroom yield. Pleurotus sapidus is one the most vital fit for human consumption having high content of protein, less content of fat, vitamins and minerals while high contents of fibers. In order to conclude the effect of various techniques of compost preparation using different levels of lime concentrations (2%, 4%, 6%, & 8%). Cotton waste was collected to examine the following phases of spawn running, pinhead formation, fruiting body and yield interval was also be evaluated to select the best compost preparation methods including wetting and boiling for the commercial production. The experiment was laid out in completely randomized design (CRD).There were two methods of compost preparation, one wetting of cotton waste and second boiling of cotton waste. It was observed boiled cotton waste with 2% lime concentration took minimum number of days to complete spawn running, while wetted cotton waste with 8% lime took maximum number of days. Similarly, boiled cotton waste with 2% lime concentration gave maximum number of pinheads and also had maximum yield. It was also observed that the number of days for spawn running increase as the lime concentration increase gradually (2, 4, 6 & 8%) and also yield decrease as the lime concentration increased. It was also observed boiled cotton waste had better results as compared to wetted cotton waste. The results showed that boiled cotton waste with 2% lime concentration was the best substrate for the cultivation of Oyster mushroom.
