Biofilm inhibition and cytotoxic analysis of synthetic and natural compounds

Biofilm formation begins with attachment of free floating microorganisms to a polymeric surface. Biofilm enhances the resistance to antibiotic compounds and human immune responses. In the present study the different extracts of leaves of Glycyrrhiza glabra and seeds of Carica papaya will be used against biofilm to check their inhibition. A series of synthetic compounds for example metal complexes and nanoparticles will be analyzed against microbial biofilm. The biofilm will develop using the tube and microtiter plate assay. The hemolytic cytotoxicity of natural and synthetic compound will be checked by using hemolytic assay method against erythrocytes. The characterization of biofilms both hydrolyzed and native will be done through phase contrast microscopy. The data will be analyzed by ANOVA under CRD. The % inhibition of S. aureus biofilm by C. papaya at 12.5% was 30.4 plus minus 0.058, 25% was 38.1 plus minus 0.100, 50% was 43.5 plus minus 0.057 and by G. glabra at 12.5% was 31.3 plus minus 0.100, 25% was 41.7 plus minus 0.100 and 50% was 44.6 plus minus 0.100. These compounds also show inhibition for E. coli C. papaya at 12.5% was 22.9 plus minus 0.231, 25% was 27.3 plus minus 0.100, 50% was 30.4 plus minus 0.100 and by G. glabra at 12.5% was 16.2 plus minus 0.057, 25% was 29.0 plus minus 0.100 and 50% was 35.2 plus minus 0.057. So, higher concentration of compound causes higher the antibacterial activity. The difference in activity of extracts was due to the solibility of antimicrobial compounds. These compounds also showed some of hemolytic activity C. papaya at 12.5% was 2.01, 25% was 2.12, 50% was 2.11 and by G. glabra at 12.5% was 3.06, 25% was 3.13 and 50% was 3.41. These natural compounds have less toxicity to erythrocytes. The synthetic compounds showed great bacteriostatic activity as well as bactericidal activity. The compounds IDAS showed maximum inhibition 78.0 plus minus 0.057 and IAP17 showed 76.8 plus minus 0.057 and IDA1 showed 74.8 plus minus 0.058 and IDA6 74.6 plus minus 0.057 while IAP2 did not show any inhibition against S. aureus. The compounds IDA4 showed maximum inhibition against E. coli i.e. 76.4 plus minus 0.057 and after this IAP12 showed 72.8 plus minus 0.100 and IAP23 71.7 plus minus 0.057 and IAP17 showed 71.4 plus minus 0.100. By the characterization of native and treated biofilms concluded that selected natural and synthetic compounds produced vast difference in morphology, functional group and amount of natural compounds. The IDA5 showed minimum cytotoxicity of 1.50. This is useful for production of new antibiotics.
