Proteomic identification of GroEl protein from whitefly

Bemisia tabaci, a whitefly, is an insect that is responsible for the transmission of Begomoviruses in a non-propagative persistent circulative manner along with other viruses while feeding on different plant species. It has a drastic effect on cotton plant by injecting number of viruses. Research has revealed that GroEL protein, which is made within the insect, interacts with viral coat protein and protects it from being digested in the insect mid gut. This protein is actually a cheperonin protein that has inducive promoter and expressed when virus gets into the insect. On the other hand, it is also stated that this protein is made from an endosymbiotic bacteria Arsenophonus nasonia that is present in the mid gut and stylet of B. tabaci. The present study is focused on this understanding about the presence of GroEL protein in virus infected or non-infected whiteflies. For this, total protein was isolated from infected whiteflies and non-infected whiteflies. Protein concentrations were checked from samples of virus infected and non-infected whiteflies and were analyzed by SDS-PAGE and LCMS technique to confirm presence and absence of GroEL protein. It showed that the protein was present in soluble protein sample of infected whiteflies. The blast results showed the similarity of protein to GroEL protein secreted by symbiotic bacteria, which is usually present in midgut of whitefly. The result has leaded to make strategies against virus control as CP of virus interacts with GroEL.
