Monitoring of formation of biofilms by opportunistic and pathogenic bacteria | Мониторинг образования биопленок условно-патогенными и патогенными бактериями

English. The aim of the work is monitoring the formation of biofilms by opportunistic and pathogenic microorganisms. The cultures of the genera Salmonella, Escherichia, Yersinia, Proteus, Citrobacter, Enterobacter, Prtovidenzia, Morganella, Klebsiella, Cronobacter, Pseudomonas, Bacillus, Staphylococcus were used in the work. The studied microorganisms were cultured in polystyrene 96-well plates. For this purpose, a daily culture of microorganisms was introduced into the wells with meat-peptone broth, having previously established a concentration of 10E4 mc/ml, and incubated for 24-96 h at temperature of 37 deg C. Then the medium with plankton cells was removed from the wells. 200 μl of filtered 0.1% crystal violet solution was poured into the wells of the plate and the plates were kept for 10-15 min at room temperature. Then dye was removed from the wells. Unbound dye was thoroughly washed with saline or distilled water. The plates were turned over on filter paper and dried. The presence and density of biomatrix (biofilm) was determined visually by the intensity of staining the surfaces of plates. Then, for the extraction of paint from the film, 200 μl of 96% ethanol was added to the wells and the optical density was measured on KFK-3KM spectrophotometer at a wavelength of 590 nm. The results of the experiments allowed asserting that within 48 h of cultivation microorganisms form a mature biofilm, which can serve as a model for studying the process of biofilm formation. Biofilm of microorganisms of different taxonomic groups differs in density. In addition, even bacteria belonging to the same genus, under the same conditions, can form a biofilm, the density of which differs by 30-60%.

Russian. Цель работы: осуществить мониторинг образования биопленок условно-патогенными и патогенными микроорганизмами. В работе использовали культуры родов Salmonella, Escherichia, Yersinia, Proteus, Citrobacter, Enterobacter, Providencia, Morganella, Klebsiella, Cronobacter, Pseudomonas, Bacillus, Staphylococcus. Изучаемые микроорганизмы культивировали в полистироловых 96-луночных планшетах. Для этого в лунки с мясо-пептонным бульоном вносили суточную культуру микроорганизмов, предварительно установив концентрацию 10E4 м.к/мл, и инкубировали в течение 24…96 ч при температуре 37 град. С. Затем удаляли из лунок среду с планктонными клетками. В лунки планшета вносили по 200 мкл отфильтрованного 0,1%-го раствора кристаллического фиолетового и выдерживали планшеты в течение 10…15 мин при комнатной температуре. Затем удаляли из лунок краситель. Не связавшийся краситель тщательно смывали физиологическим раствором или дистиллированной водой. Планшеты переворачивали на фильтровальную бумагу и высушивали. Наличие и плотность биоматрикса (биопленки) определяли визуально по интенсивности окрашивания поверхности планшета. Затем для экстракции краски из пленки в лунки добавляли по 200 мкл 96%-го этанола и измеряли оптическую плотность на спектрофотометре КФК -3КМ при длине волны 590 нм. Результаты проведенных экспериментов позволили утверждать, что в течение 48 ч культивирования микроорганизмы формируют зрелую биопленку, которая может служить моделью для изучения процесса биопленкообразования. Биопленка микроорганизмов различных таксономических групп различается по плотности. Помимо этого даже бактерии, относящиеся к одному роду, в одинаковых условиях могут образовывать биопленку, плотность которой различается на 30…60%.
