Effect of elicitors on the growth and production of monoterpenes from Michelia alba callus cultures

In this work, we attempt to monitor the effect of elicitation on callus growth and production of monoterpene compounds M.alba callus culture. A higher increased in callus growth and monoterpene production was observed after exposure to 0.2mg/ml (w/v) pectinase. However, other elicitor such as jasmonic acid (JA) failed to show any stimulatory effect on both callus growth and monoterpene production. Callus cultures treated with JA showed the low biomass accumulation and monoterpene production as compared to the cultures grown under pectinase treatment and control (supply with 25mg/l (w/v) casein hydrolysate). Cinnamaldehyde was the major compound detected in jasmonic acid and pectinase treatment, i.e, 40 and 65% respectively. A total 7 compounds was detected in the entire callus treated with 0.2mg/ml (w/v) of jasmonic acid, but there are 16 compounds were detected in callus treated with 0.2mg/ml (w/v) of pectinase. Treatment of callus culture with 0.25mg/ml (w/v) of pectinase gave the highest performance biomass accumulation such as 0.118±0.002gDWt/flask on day 18th, followed by jasmonic acid (0.2mg/ml) (w/v), 0.103±0.003g DWt/flask on day 30th. Although, callus culture under elicitation condition was unable to produce linalool, which was the major of monoterpene compounds in M.alba flower (Ueyama et al,1992).
