Поиск полиморфных вариантов гена LCORL с помощью секвенирования по Сенгеру у пород кур различного направления продуктивности | Search for polymorphic variants of the LCORL gene using Senger sequencing in chickens of various directions of productivity

Russian. Выявлено влияние полиморфных вариантов гена LCORL у многих видов с.-х. животных. Предполагается, что ген LCORL ассоциирован с размерами скелета у кур, но еще недостаточно изучен. Стоит задача найти новые полиморфные варианты в гене LCORL у генофондных пород. Цель исследования - поиск и анализ полиморфных вариантов в гене LCORL с помощью секвенирования по Сенгеру у пород различного типа продуктивности. Исследования проводили на базе лаборатории молекулярной генетики. Объектом эксперимента служили популяции 4 пород кур разного направления продуктивности биоресурсной коллекции "Генетическая коллекция редких и исчезающих пород кур": корниш, китайская шелковая, итальянская куропатчатая, пушкинская. Материалом для исследования послужил 61 образец ДНК, амплификацию проводили на приборе Thermal Cycler T100 (Bio-Rad, США). Последовательности нуклеотидов определяли на автоматическом секвенаторе Applied Biosystems 3500 (Thermo Fisher Scientific Inc., США) в лаборатории молекулярной генетики. Для секвенирования использовали набор реагентов Big Dye Terminator Cycle Sequencing Kit (Applied Biosystems, США). Биометрическая обработка данных выполнена с помощью программы Microsoft Excel. В проведенном исследовании выявлена генетическая изменчивость по полиморфным вариантам A30G, G82C, G236T, A342G, A450C, A503G, A508G в интроне гена LCORL популяций кур различного направления продуктивности. По замене A30G в популяции корниш 87% особей выборки носители генотипа АА. Совсем противоположная ситуация в породах итальянская куропатчатая, все особи – носители генотипа GG, а в китайской шелковой популяции 72% особей – носители GG. Эти породы значительно отличаются по экстерьерному профилю, таким образом представляет интерес изучение частоты встречаемости замены A30G. Полученные результаты позволяют предположить, что полиморфный вариант A30G гена LCORL можно рассматривать в качестве ДНК-маркера признака "размеры скелета" у кур.

English. The effect of polymorphic variants of the LCORL gene in many species of farm animals was revealed. It is believed that the LCORL gene is associated with skeleton sizes in chickens, but has not yet been adequately studied. The task is to find new polymorphic variants in the LCORL gene in gene pool breeds. The aim of the work is to search for and analyze polymorphic variants in the LCORL gene using Senger sequencing in breeds of various types of productivity. The studies were carried out on the basis of the laboratory of molecular genetics. The object of the experiment was populations of 4 breeds of chickens of different directions in productivity of the Biological Resource Collection "Genetic Collection of Rare and Endangered Breeds of Chickens": Kornish, Kitayskaya shelkovaya, Ital'yanskaya kuropatchataya, Pushkinskaya. 61 DNA samples served as the material for the study. Amplification was performed on a Thermal Cycler T100 instrument (Bio-Rad, USA). Nucleotide sequences were determined on an Applied Biosystems 3500 automated sequencer (Thermo Fisher Scientific Inc., USA) at the Laboratory of Molecular Genetics. The Big Dye Terminator Cycle Sequencing Kit (Applied Biosystems, USA) was used for sequencing. Biometric data processing was performed using Microsoft Excel. The study revealed genetic variation in polymorphic variants A30G, G82C, G236T, A342G, A450C, A503G, and A508G in the intron of the LCORL gene of chicken populations of different directions of productivity. As to substitution of A30G in the Kornish population there were selectd the carriers of AA genotype. Entirely observe was in the Ital'yanskaya kuropatchataya breeds, all individuals were the carriers of GG genotype, in the Kitayskaya shelkovaya population 72% of individuals were the carriers of GG genotype. These breeds greatly differ in the exterior profile, thus the study of occurrence of A30G substitution. The results suggest that the polymorphic A30G variant of the LCORL gene can be considered as a DNA marker for the sign of "skeleton size" in chickens.
