Determination of leptin, some vitamins and biochemical parameters in differentbreeds of sheep. YYU, Institute of Health Sciences MSci Thesis in Department of BiochemistryLeptin plays an important role in nutrition and energy homeostasis. Leptin is also known as a satietyfactor, which coordinates the control of body weight. Leptin hormone is produced by adipocytes. In thisstudy it was aimed to determine the levels and the correlations of leptin, some vitamins (retinol, α-tocopherol, vitamin D3) and some biochemical parameters (glucose, urea, total protein, T3) betweenNorduz and Tahirova sheep breeds in Yuzuncu Yil University Research Livestock Farm. In the samefeeding conditions two different of healthy sheep breeds, 13 each of the breeds, total of 26 sheep were useas live research materials. All of them were weighed in the morning at the beginning of July after pastureThe highest live weighted sheep were selected among them and no additional nutrients provided to allanimals throughout the trial. Then blood samples were taken from the jugular vein of all animals. Leptinanalysis were done by Cusabio brand Sheep leptin ELISA kit. Vitamin A, D and E amounts weremeasured by HPLC. The levels of glucose, urea, total protein, FT3 and FT4 were determined byautoanalyzer (Architect plus 160000). The levels of glucose, urea, leptin, total protein, FT3, FT4, retinol,α-tokoferol, Vitamin D3 in Tahirova and Norduz sheep were; 37.54-38.00 mg/dL, 32.69-34.15 mg/dL,15.92-5.22 ng/dL, 7.55-7.38 g/dL, 3.38-3.10 pg/dL, 1.07-0.91 ng/mL, 0.90-1.09 μg/mL, 1.38-1.69 μg/mLand 0.02–0.02 μg/mL, respectively. In Tahirova breed leptin and FT4 levels were higher and statisticallysignificant(p<0.001). The average values of other parameters examined such as glucose, urea, totalprotein, T3, retinol, α-tocopherol, vitamin D3 had no significant differences between in Tahirova ofNorduz sheep. A negative correlation between total protein-FT3 and alpha-tocopherol in Tahirova sheep(p<0.05, p <0.01 repectively), positive correlation between retinol-D3 (p<0.05), whereas in Norduz sheepnegative correlation between FT3-urea (p <0.01), FT4 - urea (p <0.01), retinol -leptin (p<0.05), vitamin D3-leptin (p <0.05), and positive correlation between FT4 and FT3 (p <0.05) were determined. The results ofresearch are referring to in which the serum leptin levels of thin-tailed sheep breeds detected as high, theoretically shows that of leptin production is not depending on the fat content of tail. Additional feedhormonestudies will be necessary to clarify the biochemical mechanisms of such incidents in the thin andfat-tailed sheep.
