Determination of malonaldehyde and hexanal by solid phase microextraction-gas chromatogtaphy method

In this study, the effects of matrix (air matrix and sunflower oil matrix), extraction temperature (40, 50 and 60 °C) and extraction time (20, 30 and 40 minutes) on extraction of hexanal (HEG) and malonaldehyde (MAD) by Solid Phase Micro-extraction fiber (SPME) were investigated. 5-Methyl 2-hexanone (0.5 ppm) was used as Internal Standard (IS) for quantification of HEG and MAD. In order to determine the level of linearity between HEG and MAD with IS, various ratios of HEG and MAD (5 ppb, 10 ppb 50 ppb, 100 ppb, 0.5 ppb, 1 ppm, 5 ppm and 10 ppm) and IS (0.5 ppm) were added to sunflower oil samples and empty vials, and by applying the SPME technique the relation between variables (x = Concentration of HEG or MAD/Concentration of IS, y = Area of HEG or MAD/Area of IS) were evaluated by regression equation (y=ax+b) and correlation coefficient (R2). Both HEG and MAD demonstrate a good correlation with IS in the air matrix, but because of the high reactivity and polarity of MAD, the correlation between MAD and IS was not sufficient (0.015 ˂ R2 ˂ 0.606) to explain the linearity between MAD and IS in the sunflower oil and consequently using 5-methyl 2-hexanone as IS for quantification of MAD in sunflower oil. By evaluation the R2 values between HEG and IS, it was found that the most appropriate condition for extraction of HEG was 60 °C and 20 minutes.As a result, SPME which is a simple, fast, sensitive and solvent-free technique can be used in determination of HEG in both air and sunflower oil matrix, while for determination of MAD in sunflower, it must be derivatized to analysis by SPME.
