Basal Stem Rot (BSR) disease which is caused by fungal species of Ganoderma has posted a threat to Malaysian oil palm industry. Among the identified species, the most aggressive species that caused the disease is Ganoderma boninense. It is important to have molecular information and knowledge on the fungus biochemical process in order to find the effective ways to tackle the disease. Cyclophilin-encoding (cyp) gene has been associated with fungal pathogenicity in a number of studies. Some of the studies have also indicated that deletion of the cyp gene reduced the fungal infectivity. In this study, degenerate primers have been designed based on the Cyclophilin_ABH-like conserved domain regions. First-strand cDNA was prepared from the G. boninense total RNA. SMARTer RACE PCR amplification has successfully amplified a 483 bp fragment of G. boninense cyp cDNA including 113 bp of 3' UTR region. BLASTX result showed that the sequence has about 90 % similarity to other fungal cyclophilins. The coding region codes for 113 deduced amino acid residues. Conserve domain search result showed that the sequence has some similarity to the Cyclophilin_ABH-like domain.
