Influence of nutrient medium composition on susceptibility and antagonistic properties of the nisin producer | Влияние состава питательной среды на жизнеспособность и антагонистические свойства продуцента низина

Russian. Для культивирования продуцентов низина предложена питательная среда, полученная путем смешивания в псевдокипящем слое ферментативного гидорлизата молозива коров и питательной среды MRS. Изучены антогонистические свойства, титр клеток и устойчивость к окислению продуцента низина Lactobacillus lactis, культивируемого на модифицированной питательной среде. Продуцентом низина служила односуточная культура Lactobacillus lactis АТСС 19435. Культивирование проводили на чистой среде MRS (контроль) и среде с содержанием гидролизата молозива коров в количестве 5 г, 10 г и 15 г/100 г среды MRS. Через 48 ч культивирования при введении в питательную среду MRS 5 г гидролизата молозива активность культуры увеличивалась, по отношению к контролю, на 38,1%, при введении 10 г – на 36,2%, 15 г – на 35,8%. Внесение 5 г ферментативного гидролизата молозива коров в питательную среду при культивировании Lactobacillus lactis усиливает процессы роста и размножения микробных клеток исследуемого штамма после 18 ч на 64,3%, 10 г – на 78,6%, в сравнении с контролем. Следует отметить некоторое снижение титра культуры Lactobacillus lactis при введении 15 г гидролизата через 24 ч инкубации, что объясняется особенностями биосинтеза низина. При высоких концентрациях бактериоцина процесс образования низина у лактобактерий задерживается. При введении пероксида водорода после 24 ч культивирования титр культуры при внесении в питательную среду 5 г гидролизата молозива возрастал, по сравнению с контролем, в 0,3 раза, 10 г гидролизата – в 3,3 раза, 15 г – в 3,4 раза.

English. For the cultivation of nisin producers we proposed a nutrient medium, obtained by mixing the enzymatic hydrolysate of bovine colostrum and the MRS growing medium in a pseudo-boiling layer. The antagonistic properties, cell titre and resistance to oxidation of the nisin producer Lactobacillus lactis cultivated on a modified nutrient medium were studied. The nisin producer was a one-day old culture of Lactobacillus lactis ATCC 19435 Cultivation was carried out in a pure MRS medium (control) and a medium containing bovine colostrum hydrolysate in the amount of 5 g, 10 g, and 15 g/100 g of MRS medium. In 48 h of cultivation with the introduction of 5 g of colostrum hydrolyzate into the MRS medium, the culture activity increased by 38.1% compared to the control, with the introduction of 10 g – by 36.2%, 15 g – by 35.8%. The introduction of 5 g of bovine colostrum enzymatic hydrolyzate into the nutrient medium during the Lactobacillus lactis cultivation improves the growth and reproduction processes of microbial cells of the studied strain in 18 h by 64.3%, of 10 g – by 78.6%, in comparison with the control. It should be noted that there was a slight decrease in the titre of the Lactobacillus lactis culture with the introduction of 15 g of the hydrolyzate after 24 h of incubation, which is explained by the peculiarities of nisin biosynthesis. At high concentrations of bacteriocin, the nisin formation process in lactobacilli is delayed. With the introduction of hydrogen peroxide after 24 h of cultivation, the titre of the culture increased 0.3 times when 5 g of colostrum hydrolyzate was added to the nutrient medium (comparing with the control), when 10 g of the hydrolyzate was added –3.3 times, 15 g –3.4 times.
