Features of Monarda didyma clonal micropropagation at the stage of introduction into culture in vitro | Особенности клонального микроразмножения монарды двойчатой на этапе введения в культуру in vitro

Russian. Монарда двойчатая (Monarda didyma L.) является лекарственным, пряно-ароматическим и эфиромасличным растением семейства Lamiaceae. Листья и соцветия монарды отличаются высоким содержанием эфирного масла – до 3,0%. Его наиболее ценные компоненты (тимол и карвакрол) обусловливают высокую бактерицидную, фунгицидную, противовоспалительную и антигельминтную активность, что способствует применению этого растения в медицине. Растительное сырье монарды также используют в косметологии и кулинарии. Клональное микроразмножение позволяет ускорять селекционный процесс, получать генетически однородный и оздоровленный посадочный материал, создавать коллекции in vitro ценных образцов и сортов. Цель исследования – изучение влияния состава питательной среды и типа экспланта на развитие микропобегов на первом этапе микроразмножения M. didyma in vitro. Исследования проводили в лаборатории биотехнологии в 2020–2021 гг. В результате экспериментов получены данные о морфогенезе двух типов эксплантов при их культивировании на 14-ти вариантах питательной среды Мурасиге и Скуга (МС). Анализ влияния гормонального состава среды на морфометрические показатели эксплантов показал преимущество использования в качестве регулятора роста кинетина по сравнению с 6-бензиламинопурином и тидиазуроном. Установлено, что оптимальной питательной средой на этапе введения является среда МС с 1,0 мг/л кинетина, на которой получено 2,9 микропобегов на эксплант длиной 2,7 см с 2,2–2,9 узлами. При сравнении морфогенетического потенциала двух типов эксплантов (верхушек побегов и сегментов стебля с узлом) не выявлено существенных различий по количеству побегов на эксплант и их длине. Показано, что на средах с кинетином и на безгормональной питательной среде происходило корнеобразование с частотой 42,1–60,6%. Результаты исследований являются основой для разработки методики клонального микроразмножения M. didyma.

English. Geminate horsemint (Monarda didyma L.) is a medicinal, aromatic and essential oil plant of the Lamiaceae family. The content of essential oil in the leaves and inflorescences of horsemint is rather high – up to 3%. Its most valuable components (thymol and carvacrol) cause high bactericidal, fungicidal, anti-inflammatory and anthelmintic activity, which contributes to the use of this plant in medicine. Horsemint raw materials are used in cosmetology and cooking. Clonal micropropagation allows speeding up the breeding process, obtaining genetically a homogeneous and healthy planting material, creating in vitro collections of valuable accessions and cultivars. The aim of the research was to study the influence of the culture medium composition and explants type on the development of microshoots at the first stage of M. didyma L. micropropagation in vitro. The studies were carried out in 2020–2021 in the Laboratory of Biotechnology. The experiments have resulted in obtaining data on the morphogenesis of two explant types during their cultivation on 14 variants of Murashige and Skoog (MS) culture medium. An analysis of the effect of the medium hormonal composition on the explants morphometric parameters showed the advantage of using kinetin as growth regulators compared to 6-benzylaminopurine and thidiazuron. It has been established that the optimal culture medium at the introduction stage is MS medium containing 1 mg/L of kinetin, in which 2.9 microshoots per explant of 2.7 cm long having of 2.2-2.9 nodes were produced. When comparing the morphogenetic potential of two explant types (shoot tips and node stem segments), no significant differences in the number of shoots per explant and their length were found. It was shown that on media with kinetin and on a hormone-free culture medium, root formation occurred with a frequency of 42.1-60.6%. The research results are the basis for the development of a procedure for M. didyma clonal micropropagation.
