Optimum cultural conditions for Mucor pusillus polygalacturonase production

The effect of sugar concentration, inoculum size, initial pH, nitrogen source, pectin concentration and incubation time on Mucor pusillus polygalacturonase (MPPG) production in supplemented date extract medium were studied. Ammonium sulfate was the most efficient nitrogen source compared with urea, ammonium chloride, ammonium nitrate and ammonium di-hydrogen phosphate. The optimum conditions for MPPG production were 1% sugar, 0.5% pectin, 0.3% (NH4)2SO4, 0.32% MgSO4 and 0.25% KH2PO4 at an initial PH of 4.0 after 96 hrs. of incubation at 30 deg C.
