Determination of protein content in seed meal by pulsed nuclear magnetic resonance spectroscopy

This paper presents a method for determination of protein content by pulsed NMR. The research consists in the choice of the pulsed NMR program necessary for determining the time of tranversal relaxation (T2) and in the determination of the optimum parameters for each operation implied by the method. Using this technique, calibration curves for sunflower, soybean, rape seed meal, corn, castor oil plant germs were drawn and the accuracy and reproducibility of each meal under study were computed. Square mean errors of the mean values as compared to Kjeldahl methods presented minimum values of + 0.42 for corn germ meal and maximum values of + 0.87 for sunflower seed meal. The results lead to the conclusion that the method for determination of protein content by pulsed NMR offers some advantages in comparison with Kjeldahl method in respect of rapidity (duration of analysis of about 1 1/2 h as compared with 7-8 h for Kjeldahl method), simplicity of the technique and lack of reagent toxicity. As for accuracy and the repetability of the results, the method is less accurate in comparison with Kjeldahl method, the square mean error of the mean value as compared to the latter being of +/- 1%
