Effect of pentothal on rat serum amino acid pattern and T3 and T4 concentration

Since barbiturates can influence the pattern of amino acid in serum and considering tyrosine as the starting material for T3 and T4 production, a study was carried out in rats using sodium pentothal administered intra peritoneally at a dose of 50 mg/kg of body weight. The serum levels of T3 and T4 were determined by radioimmunoassay, while the free amino acids were analyzed by HPLC. Reversed phase HPLC was used, employing precolumn derivatization with o-phtaldialdehyde and ethanethiol as reducing agents, to detect the serum amino acids. The analyses of serum show that there was a significant increase in the concentration of most serum amino acids of treated animals except for arginine, valine and methionine, which remained unchanged. The greatest rise was found for the concentration of amino acids brought about significant changes in the concentration ratio individual amino acids and that of the amino acid pool. Increased ratio were noted with isoleucine, tryptophan, phenylalanine and serin. A decreased in ratio was registered for arginine, aspartic acid and tyrosine, while glutamic acid, asparagine, glutamine, glycine, alanine, arginine and methionine showed no change in their amino acid concentration toward the concentration of amino acid pool. The decreased in the concentration ratio value of tyrosine, would result in the lower concentrations of T3 and T4 found in the treated animals.
