Effects of plant growth regulator on the formation and growth callus of Solanum laciniatum using the cultures in vitro

A study was conducted at the Laboratory of Balitro. The objective was to evaluate the effects of auxin (2,4-D), cytokinin (BAP, kinetin) and the coconut water on callus formation of the plant's tissues of Solanum laciniatum Ait., in the in vitro culture. The treatment consisted of 11 kinds of media, those were the combination of 3 and 4 mg/12,4-D with 0,5 mg/l BAP or kinetin or 15 percent of the coconut water. Explant used is leaf derived from plantlet in a sterile bottle. The design was completely randomized. The first sub culture was carried out at the age of 60 days passage 1 and second culture as passage 2. While at the second passage medium was treated with combination of 1-5 mg/l 2,4-D and 0,5 mg/l BAP or kinetin. Parameters assessed were callus formation, fresh weight of callus at 30 and 60 days, dry weight of callus, structure and colour of callus and solasodin content. From the experiments it was revealed that all treatments produced similar responses to callus formation. The medium MURASHIGE and SKOOG of explant S. laciniatum did not form any plantlet, but a few callus. Cytokinin application either BAP or kinetin on the medium containing 2,4-D was able to accelerate a callus formation and increase the fresh weight of callus. Application of coconut water also accelerated callus formation and increased fresh weight of the callus. BAP and kinetin increased the weight of callus after 30 days. BAP was more effective compared with kinetin on increasing fresh weight of callus. The average solasodine content produced by various treatments of callus of S. laciniatum was 0,2 percent dry weight.
