Monoclonal antibodies reactive to some Pseudomonas fluorescens proteases [raw milk]

The production is described of monoclonal antibodies directed against the spoilage protease secreted by a strain of Pseudomonas fluorescens isolated from raw milk, and their use in an enzyme-linked immunosorbent assay (ELISA) to quantify this enzyme. Some P. fluorescens proteases have previously been shown, using polyclonal antisera, to be immunologically dissimilar; however, in this work one protease was used as the immunising antigen and the resulting monoclonal antibody cross-reacted with these other, dissimilar enzymes. The antibody was used to measure P. fluorescens protease in artificially contaminated, ultra heat treated (UHT) milk by a competitive ELISA by which as little as 3.1 ng of enzyme per mL of milk could be detected.
