Serum alkaline phosphatase activity isoenzymes in ewes and their lambs

English. We studied the total activity and the types of isoenzymes of alkaline phosphatase in sera of ewes during the last month of pregnancy, in the same animals three-four weeks after lambing and in sera lamb at four weeks of age. Mean total SAP activities were 224.9 +- 81.3 Iu/l for the pregnant ewes, 196.8 +- 84.7 IU/L post-partum and 1573.6 +- 643.6 IU/L for the lambs. For identification of different isoenzymes we employed agarose electrophoresis of fresh serum, after heating at 56 deg C and after treatment with neuraminidase. In ewes, two isoenzymes were identified (skeletal and hepatic). There was no difference in the banding patterns of pregnant and post-partum sheep. In lamb serum there were found three isoenzymes, skeletal, intestinal and a third of uncertain origin. To identify this third isoenzyme, it must be looked for intestinal as well as hepatic homogenates

Italian. L'attivita' sierica dell'ALP e dei suoi isoenzimi e' stata studiata in un gruppo di pecore al termine della gravidanza e all'inizio della lattazione, nonche' nei loro agnelli. Considerando anche le eventuali prospettive diagnostiche, si sono adottate metodiche di agevole impiego: per l'identificazione dei diversi isoenzimi si e' ricorsi alla separazione elettroforetica su gel d'agarosio condotta su siero t.q., dopo inattivazione termica a 56 gradi C per 10 minuti e dopo trattamento con neuraminidasi. Si e' cosi' potuta individuare nelle pecore la presenza di due bande, di cui una identificabile con l'isoenzima osseo e l'altra verosimilmente con quello epatico. Nessuna differenza e' stata riscontrata nei due momenti fisiologici considerati. Negli agnelli sono risultate presenti tre bande, di cui la piu' evidente di origine scheletrica, una seconda di probabile origine intestinale e una terza la cui attribuzione e' attualmente incerta e potra' essere ben definita attraverso ulteriori indagini su omogenati d'organo
