[Effect of pulse treatment with NAA and IBA on root formation of microcuttings of vanilla (Vanilla planifolia Andr.)]

Successful in vitro shoot multiplication of vanilla (Vanilla planifolia Andr.) has been reported, but root formation of vanilla microcuttings is still not clear. The objective of this experiment was to investigate the effects of pulse treatment with NAA (Naphtaleneacetic Acid) and IBA (Indolebutyric Acid) on root formation of microcuttings of vanilla. The experiment was arranged in a completely randomized design with 20 replications. The microcuttings were incubated for 2 weeks in agar-solidified medium containing half-strength MS salts, 3 sucrose, 100 mg/l myo-inositol, 0.5 mg/l pyridoxine.HCl, 0.5/l nicotinic acid, 0.1 thiamine.HCl and 300 M IBA or 300 M depending on the treatments, and then cultured for 4 weeks on the same medium without plant growth regulators. The results showed that neither NAA nor IBA induced adventitious root formation. Eight of 32 microcuttings planted on soil medium formed aerial roots. Therefore, it is recommended that it is not necessary for microcuttings to have adventitious roots in order to survive ex vitro. Instead, the multiplication stage should produce microcuttings which can form aerial roots ex vitro
