Chromosomal distribution of sequences homologous to the Bg transposable element in maize (Zea mays L.) inbred lines

English. The chromosomal locations of sequences homologous to the Bg element were determined in four maize inbred lines. Two linkage maps obtained from segregating populations, derived from A7 x B73 and Lo881 x Lo951 crosses respectively, were used. A total of 58 polymorphic fragments were mapped with the use of two restriction enzymes (EcoRI and HindIII) and two Bg probes. Localization of the polymorphic bands on the two maps resulted in the identification of 35 independent loci. Bg-like sequences were revealed on nine of the maize chromosomes. They were present in 6-7 chromosomes in the inbred lines Lo881, Lo951 and B73, while A7 hosted Bg-like sequences in only four chromosomes. Although the positions of homologous sequences varied in different inbreds, the majority of Bg-homologous sequences were found at linked positions. The distribution pattern of Bg-like sequences was thus in agreement with transposition of the element to linked sites. The highest number of Bg-like sequences was localized on chromosome seven. Analyses to evaluate the presence, amongst the inbred lines, of deletion derivatives of the Bg element indicated that such sequences are virtually absent. Considering the results presented and earlier data demonstrating that Bg-like sequences are present at a frequency of 8.7 +- 2.1 among inbred lines, we estimated that the loci which were mapped represent not less than 80% of the Bg-like sequences present in the genome of the inbreds

Italian. [In quattro linee consanguinee di mais sono state determinate le localizzazioni delle sequenze omologhe all'elemento Bg. Sono state utilizzate due mappe di concatenazione ottenute da popolazioni segreganti derivate, rispettivamente, dagli incroci A7 x B73 e Lo881 x Lo951. Con l'impiego di due enzimi di restrizione (EcoRI e HindIII) e due sonde Bg e' stato mappato un totale di 58 frammenti polimorfici. La localizzazione delle bande polimorfiche sulle due mappe si e' tradotta nell'identificazione di 35 loci indipendenti. In nove dei cromosomi del mais sono state evidenziate sequenze Bg-simili. Esse erano presenti in 6-7 cromosomi nelle linee consanguinee Lo881, Lo951 e B73, mentre A7 presentava sequenze Bg-simili in solo 4 cromosomi. Sebbene le posizioni delle sequenze omologhe variassero nelle diverse linee consanguinee, la maggioranza delle sequenze dei Bg-omologhi sono state ritrovate nelle posizioni concatenate. Il tipo di distribuzione delle sequenze Bg-simili era in tal modo in accordo con la trasposizione dell'elemento ai siti concatenati. Il maggior numero delle sequenze Bg-simili era localizzato sul cromosoma 7. Le analisi per valutare la presenza, fra le linee consanguinee, di derivati di delezione sull'elemento Bg indicavano che tali sequenze sono virtualmente assenti.
