Influx and efflux kinetics of ammonium transport in maize roots [Zea mays]

English. Influx of sup(13)NH4+ at the plasma membrane was measured for roots of intact seedilngs of maize (Zea mays L.) as a function of the external NH4+ concentration. The results revealed a saturable, high affinity transport mechanism (HATS) in the concentration range below 1.0 mM with a Km of 150 micronM. At external concentrations higher than 1.0 mM, influx followed a linear relation, indicating operation of a low affinity transport mechanism (LATS). Prior N-starvation decreased the Km for HATS, but reduced influx via LATS. Efflux analysis of sup(13)N, indicated cell wall and cytoplasmic compartments with half-times of exchange of 30 sec and 14 min, respectively. Estimates are given of the concentrations of NH4+ in these compartments. It is shown that influx exceeds efflux by a factor of nearly 5 under our conditions

I risultati hanno evidenziato l'esistenza di un meccanismo di trasporto ad alta affinita' saturabile (HATS) nell'intervallo di concentrazione al di sotto di 1.0 mM, con una Km di 150 micronM. A concentrazioni esterne maggiori di 1.0 mM, l'influsso seguiva una relazione lineare, indicando l'esistenza di un meccanismo di trasporto a bassa affinita' (LATS). Una precedente carenza di N determinava una diminuzione della Km per HATS, ma riduceva l'influsso via LATS.
