The setting-up of PCR-RAPD techniques for the identification of olive cultivars was achieved in this study. According to our results RAPD markers seem to be reliable when both strict protocol and conservative interpretation of data analysis is followed. Fifty-one olive (Olea europaea L.) cultivars from the Germplasm Bank of Cordoba were screened using RAPD markers. A high degree of polymorfism was evidenced in the olive germplasm studied. Forty-six out of ninety-five primers screened were selected. They provided a hundred and ninety reproductible amplification fragments from which a hundred and twenty were useful as polymorfic markers. All the cultivars studied were identified with four primers (OPA-01; OPK-08; OPX-01 Y OPX-03). The study also succeeded in identifying cultivars from samples provided by nurseries. Cluster analysis of both cultivars and zones revealed same degree of relationship between the olive cultivars and their geographic origin. A higher level of polymorfism was observed when poliacrilamide matrix was used to separate the amplification products. Poliacrilamide offers more security in data interpretation and more possibilities for cultivars' discrimination as the number of unique markers was considerably increased.
