Rapid genotypic differentiation of Lactococcus lactis subspecies and biovar

A highly efficient, rapid and reliable PCR-based method for distinguishing the subspecies and biovar of Lactococcus lactis is described. Primers complementary to positions in the histidine biosynthesis operon with distinct specificities have been constructed. PCR analysis with extracted DNA or with cells of different Lactococcus lactis strains resulted in specific fragments which could be visualized after agarose gel electrophoresis. The lenght polymorphisms of the PCR fragments allowed a clear distinction of the L. lactis subspecies and biovar. In the study, we have also included strains which in contrast to their 16S rRNA sequence belong to a different subspecies as determined by their phenotype. The results of the PCR assay agree perfectly well with the identification based on 16S rRNA analysis.
