The effects of extracellular calmodulin of style and pollen on pollen germination and pollen tube growth

The roles of extracellular calmodulin (CaM) of style and pollen in regulating pollen germination and tube growth were investigated by semivivo method. Pollen germination on stigma was inhibited when the stigma was pretreated with EGTA (Figs. 1, 2) or anti-CaM serum (Figs. 3, 4). Pollen tube growth in style transmitting tissue was accelerated when the style was microinjected with exogenous purified CaM (Fig. 5), but was inhibited by microinjection with anti-CaM serum (Fig. 6). Elisa and biotinylated CaM overlay analyses showed that CaM and calmodulin-binding proteins(CaMBPs) (Fig. 7) were present in both pollen and style cell wall, and the kinds of CaMBPs in pollen cell wall were different from those in style cell wall. The above results suggest that the extracelllular CaM of both style and pollen was able to stimulate pollen germination in the stigma and pollen tube growth in the transmitting tissue of the style
