Instability of transgene expression in lettuce (Lactuca sativa): possible causes and prevention

Transgene silencing is a major factor limiting the genetic engineering of lettuce. Although lettuce is relatively easy to transform, transgene expression is often low and transgenes are poorly expressed in subsequent seed generations. Possible factors have been investigated which may contribute to this, including transgene copy number, the promoter used to direct transgene expression and methylation. The principal strategies to increase the frequency of high, stable transgene exprcssion in lettuce include rigorous screening for plants with single, high expressing transgene inserts, and the use of plant gene promoters, such as the -784 bp petE (truncated pea plastocyanin promoter) and ACT1 (Arabidopsis actin promoter), instead of viral gene promoters, such as CaMV35S, to direct transgene expression.
