Rice, Oryxa sativa growth and final grain are limited by chilling temperatures which may occur during the tillering stage. Low temperature sensitivity of plants and algae is related to the physical state of membrane lipids which partly depends on the degree of unsaturation of membrane fatty acids. It has been established that the fad7 gene, which encodes a chloroplastic omega-3 fatty acid desaturase, is responsible for the TA 16:3 and 18:3 formation in leaf tissues. Fad7 cDNA,m screened from A. thaliana (acquired from Prof. Chris Somerville), has been introduced to rice immature embryos using microprojectile bombardment, ACCELL TM. The cloning strategy of the tranformation vector, involved the following. The gene encoding sequence was placed downstream of the ubiquitin constitutive promoter which is highly expressed in rice.
