Gametoclonal variation in anther culture-derived rice plants. 2: Segregation of mutated plants at the first progeny generation [Oryza sativa L.]

Gametoclonal variation is frequently observed in rice plants derived from anther culture, when mutant lines in which mutated plants segregate (segregating) or mutant lines in which the same mutant phenotypes are shown (non-segregating) appear. To determine which segregating mutant lines or non-segregating mutant lines appear more frequently, 108 doubled haploid (DH) lines derived from the anther culture of three inbred japonica rice cultivars, Notohikari, Akihikari and Koshihikari, were generated and their phenotypic changes were investigated in the first selfed progeny (A1) generation. Mutated plants obviously segregated in 20 DH lines. The mean and/or variance in at least one of four quantitative traits, culm length, panicle length, spikelet number and seed fertility, in 32 DH lines significantly changed compared with these traits in the seed-derived original cultivars, although the mutated traits showed a continuous distribution. Of the 32 DH lines, 14 lines showed a small variance and 18 lines had a significantly larger variance than the seed-derived control lines, indicating that the latter lines were segregating. In total, 52 of 108 DH lines (49%) had mutated genes, of which 38 (73%) were segregating lines and 14 (27%) were non-segregating lines

[In piante di riso derivate da coltura di antere, sia quando compaiono linee mutanti in cui le piante mutate hanno segregato (segreganti), sia quando compaiono gli stessi fenotipi mutanti (non segreganti), si osserva di frequente variazione gametoclonale. Per determinare quali compaiono piu' frequentemente tra le linee mutanti non segreganti e quelle segreganti, sono state generate 108 linee doppio aploidi (DH) derivate da coltura di antere di tre cultivar di riso japonica autofecondate, Notohikari, Akihikari e Koshihikari, e sono stati studiati i loro cambiamenti fenotipici nella prima generazione autofecondata della progenie (A1). Le piante mutate hanno ovviamente segregato in 20 linee DH. La media e/o la varianza in almeno uno dei quattro caratteri quantitativi, lunghezza del culmo, lunghezza della pannocchia, numero delle spighette e fertilita' del seme, in 32 linee DH cambiava significativamente rispetto agli stessi caratteri nelle cultivar originali derivate da seme, sebbene i caratteri mutati presentassero una distribuzione continua.
