Cytokines in milk from mastitic and non-mastitic cows

English. The expression of several cytokines (IL-1alpha, IL-1beta, IL-2, IL-4, IL-6, IL-8, IL-10, IL-12 p40, TNFalpha, TNFbeta, IFNgamma) has been detected by reverse transcriptase-polymerase chain reaction (RT-PCR) in milk cells collected from 24 cows (12 healthy and 12 with naturally occurring mastitis). Differences in cytokine profiles were found in the two groups of cows. In particular, mostly regulatory cytokines (IL-4, IL-10, IFNgamma) were detected in milk samples obtained from normal cows. In contrast, both regulatory and proinflammatory cytokines (IL-1alpha, IL-10, IL-6, IL-8, IL-12 p40, TNFalpha, TNFbeta) were found in milk from naturally occurring mastitis. It is quite interesting to note that two proinflammatory cytokines (IL-1beta and IL-8) were also detected in some healthy control cows, while IL-1alpha, IL-6, IL-12 e TNFalpha were detected exclusively in milk from cows with naturally occurring mastitis. No regulatory cytokine IL-2, pivotal molecule in cytokine network, was detected in all examined samples. This study shows that milk cytokine profiles could lead to a better understanding of some ubiquitous cytokines (IL-1beta, IL-8, IL-4, IL-10, IFNgamma) and it may provide, by using a quantitative analysis, an useful aid for their potential use for diagnosis, prophylaxis and immune therapy of mastitis in cows

Tramite reverse transcriptase-polymerase chain reaction (RT-PCR), e' stata ricercata l'espressione di alcune citochine (IL-1alfa, IL-1beta, IL-2, IL-4, IL-6, IL-8, IL-10, IL-12 p40, TNFalfa, TNFbeta, IFNgamma) nelle cellule del latte di 24 bovine. I risultati hanno dimostrato la differenza nei profili citochinici tra vacche sane (n. 12) e vacche con mastite (n. 12). In particolare, nei campioni da vacche sane sono state ritrovate prevalentemente citochine regolatorie (IL-4, IL-10, IFNgamma), mentre nel latte di bovine con mastite sono state individuate citochine sia regolatorie, sia proinfiammatorie (IL-1alfa, IL-1beta, IL-6, IL-8, IL-12 p40, TNFalfa, TNFbeta). E' interessante notare come 2 citochine proinfiammatorie (IL-1beta e IL-8) sono state identificate anche in alcuni soggetti sani, mentre IL-1alfa, IL-6, IL-12 e TNFalfa sono esclusive delle bovine con mastite. Inoltre, segnaliamo che l'IL-2, molecola pivotale nel network citochinico, non e' stata ritrovata in alcun campione. Riteniamo, infine, che questi risultati offrano un contributo all'individuazione di quelle citochine ubiquitarie (IL-1beta, IL-8, IL-4, IL-10, IFNgamma) la cui determinazione quantitativa potrebbe essere utile allo sviluppo di strategie immunomodulatorie nella profilassi e terapia delle mastiti
