Bacterial expressed coat protein. Development of a single antiserum for routine detection of Citrus tristeza virus

English. Citrus tristeza virus (CTV) causes one of the most important citrus diseases worldwide and has recently been detected in Portugal. Early diagnosis based on immunoenzymatic techniques requires significant amounts of reagents. This research describes the isolation of the coat protein gene of CTV, its expression in Escherichia coli as a fusion protein containing an N-terminal (His) sub (6) region and its use to raise rabbit polyclonal antibodies. These antibodies could be used at dilutions higher than 1/80,000 at the detection stage in indirect ELISA tests and were also efficient for trapping the virus in ELISA and Immunocapture RT-PCR. These characteristics allowed the production of diagnostic kits based solely on this source of antibodies. A detection spectrum and sensitivity similar to those of a commercial polyclonal antibody kit was achieved

Italian. [Il Virus della tristeza degli agrumi (CTV) causa una delle malattie piu' importanti degli agrumi a livello mondiale ed e' stato scoperto di recente in Portogallo. La diagnosi precoce basata su tecniche immunoenzimatiche richiede quantita' significative di reagenti. Questa ricerca descrive l'isolamento del gene della proteina capsidica di CTV, la sua espressione in Escherichia coli come proteina di fusione contenente una regione N terminale (His) inf (6) e la sua utilizzazione per sviluppare anticorpi policlonali nel coniglio. Questi anticorpi potevano essere utilizzati a diluizioni maggiori di 1/80.000 nello stadio di diagnosi in test ELISA indiretti ed erano pure efficaci per intrappolare il virus nell'ELISA e nella RT-PCR con Immunocattura. Queste caratteristiche hanno consentito la produzione di kit diagnostici basati solamente su questa fonte di anticorpi.
