The using of gamma-octanolactone in the broiler diet: intestinal microbiome | Использование гамма-октанолактона в рационе бройлеров: микробиом кишечника

Gamma-lactones are natural compounds that have antibacterial, fungicidal, antifeedant, growth-regulating, insecticidal, attractant, and antitumor activity and can be used as an alternative to antibiotics. Gamma-octalactone was isolated from an Eucalyptus viminalis extract. Its effect on colonization of the intestine with pathogenic microflora was studied when they were added to the diet of broiler chickens. The ileum microbiome was evaluated by sequencing. Total DNA from samples of intestinal contents was isolated using a FastDNA SPIN Kit for Faeces. During the experiment, broiler chickens were divided into 3 groups of 30 heads each. The control group received only the basic diet, the 1st experimental group received gamma-octalactone in a dose of 0.05 to the basic diet, and the 2nd group received 0.20 ml/kg of live weight/day. When analyzing the bacterial profile of the samples of the ileum contents of broiler chickens of the control group 4 phyla of eubacteria were isolated, in chickens of the 1st group - 8 phyla, in chickens of the 2nd group - 5 phyla. The Firmicutes phylum was determined to be dominant in all groups, whose share in the groups was 86.1; 93.13; 94.55%, respectively. The Simpson and Shannon indices were used in the analysis of biological diversity. The Simpson index indicates the dominance of one or more bacterial species; in the control group chickens it was 0.08; 2-nd group - 0.22; 1st group - 0.30. The Shannon Index reflects the complexity of the structure of the cenosis, the greater its diversity, the greater the index. The groups are distributed in the following order: control - 3.15; 2-nd group - 2.18; 1-st group - 2.15. The results obtained allow concluding that the chickens of the experimental groups showed an increase in the number of representatives of the Firmicutes phylum; chickens of the 1st group - a decrease in the content of the genus Streptococcus by 4.21%; in chickens of the 2-nd group - an increase in representatives of the Lactobacillus and Turicibacter genera by 10% relative to the control group.

Гамма-лактоны - натуральные соединения, которые обладают антибактериальной, фунгицидной, антифидантной, рострегулирующей, инсектицидной, аттрактантной, противоопухолевой активностью и могут использоваться в качестве альтернативы антибиотикам. Гамма-октанолактон был выделен из экстракта Eucalyptus viminalis. Исследовали его влияние на заселение кишечника патогенной микрофлорой при добавлении в рацион цыплят-бройлеров. Оценку микробиома подвздошного отдела кишечника осуществляли методом секвенирования. Тотальную ДНК из образцов содержимого кишечника выделяли при помощи набора FastDNA SPIN Kit for Faeces.В ходе эксперимента цыплят-бройлеров разделили на 3 гр. по 30 гол. в каждой. Контрольная группа получала только основной рацион, 1-я опытная гр. к основному рациону получала гамма-октанолактон в дозе 0,05, 2-я гр. - 0,20 мл/кг живой массы/сут. При анализе бактериального профиля образцов содержимого подвздошной кишки цыплят-бройлеров контрольной группы было выделено 4 филума эубактерий, у цыплят 1-й гр. - 8 филумов, у цыплят 2-й гр. - 5 филумов. Доминирующим во всех группах был определен филум Firmicutes, доля которого в группах составила 86,1; 93,13; 94,55% соответственно. При анализе биологического разнообразия были использованы индексы Симпсона и Шеннона. Индекс Симпсона указывает на доминирование одного или нескольких видов бактерий; у цыплят контрольной группы он составил 0,08; 2-й гр. - 0,22; 1-й гр. - 0,30. Индекс Шеннона отображает сложность структуры ценоза, чем больше его разнообразие, тем больше индекс. Группы распределяются в следующем порядке: контроль - 3,15; 2-я гр. - 2,18; 1-я гр. - 2,15. Полученные результаты позволяют сделать вывод, что у цыплят опытных групп отмечен рост численности представителей филума Firmicutes; у цыплят 1-й гр.- снижение содержания рода Streptococcus на 4,21%; у цыплят 2-й гр. - увеличение представителей родов Lactobacillus и Turicibacter на 10%относительно контрольной группы.
