Change of culture medium positively influences the development and quality of in vitro cattle embryos | Смена среды культивирования положительно влияет на развитие и качество эмбрионов крупного рогатого скота in vitro

English. Despite the fact that the number of embryos of cattle obtained in vitro (IVP) is increasing annually in the world, their quality is still inferior to the quality of embryos obtained in vivo. In the present work, the dependence of IVP embryos development and their quality on the conditions of renewal of the in vitro culture medium (IVC) and its volume is studied. Post mortem cow oocytes were cultured in the maturation medium, fertilized in vitro frozen-thawed sperm and transferred to a BO-VC medium with a volume of 500 or 100 mcl for embryonic development. 3 incubation variants were compared: no medium change (NMC), with complete medium change (CMC) and with partial medium change (PMC). In the first case, the embryos developed during the entire period (8 days) without updating the culture medium, in the second one - after 3 days IVC embryos were transferred for further development into drops of fresh medium, in the PMC variant, after 3 days of incubation, half an initial volume of the medium was removed and replaced with an equivalent volume of fresh medium. The results showed that compared to continuous cultivation PMC provided a significant increase in the frequency of blastocyst formation (from 23.0±1.5 and 25.8±0.8 to 45.7±4.8 and 52.1±4.9%), a decrease in the frequency of apoptosis in embryos (from 6.53±0.88 and 6.47±0.66 to 3.60±0.12 and 3.50±0.29%) and an increase in the yield of hatched blastocysts from the total the number of fertilized oocytes (from 14.9±1.5 and 11.6±3.3 to 25.2±3.9 and 40.8±3.2), regardless of the volume of the IVC medium in which the embryos developed (data are presented for the medium volume, respectively, 500 and 100 mcl), and also increased the number of nuclei in blastocysts in the case of culturing embryos in a medium having a volume of 100 mcl. Cultivation involving PMC was also more effective in basic indicators than that without medium change. The frequency of blastocyst formation (up to 44.9±0.7 and 44.1±5.0%) increased similarly to what was observed with CMC. There were no significant differences between the variants of CMC and PMC across the entire spectrum of the studied indicators.

Russian. Несмотря на то, что число эмбрионов крупного рогатого скота (КРС), получаемых in vitro (IVP), в мире ежегодно возрастает, их качество все еще уступает качеству эмбрионов, получаемых in vivo. В представленной работе изучена зависимость развития IVP эмбрионов и их качества от условий обновления среды in vitro культивирования (IVC) и ее объема. Post mortem ооциты коров культивировали в среде созревания, оплодотворяли in vitro заморожено-оттаянной спермой и переносили в среду BO-IVC объемом 500 или 100 мкл для эмбрионального развития. Сравнивали 3 варианта инкубации: без смены среды (БСС), с полной сменой среды (ПСС) и с частичной сменой среды (ЧСС). В первом случае эмбрионы развивались в течение всего периода (8 сут) без обновления среды культивирования, во втором - через 3 сут IVC эмбрионы были перенесены для дальнейшего развития в капли свежей среды, в варианте ЧСС через 3 сут инкубации половина изначального объема среды удалялась и заменялась эквивалентным объемом свежей среды. Результаты показали, что по сравнению с непрерывным культивированием ПСС обеспечивала значительное повышение частоты образования бластоцист (с 23,0±1,5 и 25,8±0,8 до 45,7±4,8 и 52,1±4,9%), снижение частоты апоптоза в эмбрионах (с 6,53±0,88 и 6,47±0,66 до 3,60±0,12 и 3,50±0,29%) и повышение выхода вылупившихся бластоцист от общего числа оплодотворенных ооцитов (с 14,9±1,5 и 11,6±3,3 до 25,2±3,9 и 40,8 ±3,2) независимо от объема IVC среды, в котором происходило развитие эмбрионов (данные представлены для объема среды соответственно 500 и 100 мкл), а также увеличивало число ядер в бластоцистах в случае культивирования эмбрионов в среде объемом 100 мкл. Культивирование, предполагающее ЧСС, также было по основным показателям эффективнее, чем таковое без смены среды. Частота образования бластоцист (до 44,9±0,7 и 44,1±5,0%) повышалась аналогично тому, что наблюдалось при ПСС. Значимых различий между вариантами ПСС и ЧСС по всему спектру исследуемых показателей обнаружено не было.
