The study of the physico-chemical, the biological properties and the peptide composition of the pepsin hydrolysate of cow's colostrum | Исследование физико-химических и биологических свойств, пептидного состава пепсинового гидролизата молозива коров

English. Peptides are biologically active substances with different spectrum of action. Studies of the physicochemical properties of pepsin hydrolyzate of cow colostrum have been carried out. The enzymatic hydrolyzate of bovine colostrum has a high protein content of 9.16% and a low fat content of 0.25%. A precipitate of the hydrolyzate was obtained by centrifugation at 3900 rpm for 10 min. Two fractions R (1) and R (2) were isolated from the sediment from the pepsin hydrolyzate of cow colostrum on a MALDI-TOF mass spectrometer. It has been established that the peptide chain R (1), consisting of 6 amino acid residues in different sequences, forms a polypeptide of 11 amino acids. Peptide R (2) refers to polypeptides: its chain includes 7 repeating amino acid residues. The molecular weight of the peptide R (1) is 1.7 kDa, the peptide R (2) – 7 kDa. According to the scientific database NCBI, peptide R (1) refers to the little-studied peptide "Nuclear receptor 2C2-associated protein, Bos taurus", peptide R (2) refers to peptides "14 kDa phosphohistidine phosphatase, Pongo abelii" whose biological functions have not been studied. When studying the cytotoxicity of the isolated peptides in the composition of the bovine colostrum hydrolyzate on HEG 293 cells using the Neutral Red Assay method with a 24-hour incubation, it was found that the cytotoxicity of the studied hydrolyzate at concentrations from 0.1 to 0.5 mg/ml ranges from 0.9 to –6.30, which indicates its absence. The antimicrobial and antimicrobial activity of cow colostrum hydrolyzate against Escherichia coli, Bacillus subtilis and Candida albicans has been proven. Probably, the isolated peptides can be used as part of pharmaceuticals or functional products.

Russian. Пептиды относятся к биологически активным веществам различного спектра действия. Проведены исследования физико-химических свойств пепсинового гидролизата молозива коров. Ферментативный гидролизат молозива коров отличается высоким содержанием белка 9,16% и низким содержанием жира 0,25%. Получен осадок гидролизата с помощью центрифугирования при 3900 об/мин в течение 10 минут. Из осадка из пепсинового гидролизата молозива коров на МАЛДИ-ТОФ масс-спектрометре выделено две фракции R (1) и R (2). Установлено, что пептидная цепь R (1) состоит из 6 аминокислотных остатков в разной последовательности и образует полипептид из 11 аминокислот. Пептид R (2) относится к полипептидам, так как в его цепь входят 7 повторяющихся аминокислотных остатков. Молекулярная масса пептида R (1) составляет 1,7 кДа, пептида R (2) – 7 кДа. Согласно научной базе данных NCBI пептид R (1) относится к малоизученному пептиду "Nuclear receptor 2C2-associated protein, Bos taurus", пептид R (2) относится к пептидам "14 kDa phosphohistidine phosphatase, Pongo abelii", биологические функции которого не исследованы. При изучении цитотоксичности выделенных пептидов в составе гидролизата молозива коров на клетках линии HEG 293 методом Neutral Red Assay при 24 часовой инкубации установлено, что цитотоксичность исследуемого гидролизата в концентрациях от 0,1 до 0,5 мг/мл составляет от 0,9 до – 6,30, что свидетельствует об ее отсутствии. Доказана антимикробная и противомикробная активность гидролизата молозива коров по отношению к Escherichia coli, Bacillus subtilis и Candida albicans. Вероятно, выделенные пептиды могут быть использованы в составе фармпрепратов или функциональных продуктов.
