Can future type I diabetes be predicted? A study in families of affected children

The risk of developing insulin-dependent (type I) diabetes in siblings of affected children is examined, relative to HLA genotype (a risk factor attributing susceptibility to this disease due to HLA-linkage). Subjects in 160 families (diabetic probands) were studied, including 288 siblings (154 brothers 134 sisters) who were grouped together according to the number of HLA haplotypes in common with their probands. It was determined that HLA-identical siblings (both haplotypes in common) have about a 100 times greater risk of type I development than the general population, and a risk significantly greater than haplo-identical siblings (1 haplotype in common). These results indicate that 30% of HLA identical siblings in northern Europe should develop diabetes by age 30. (wz)
