Studies on the Genetic Diversity of Wheat Genotypes (Triticum durum L.) in Response to Drought Stress Through Isozyme Markers

Total eleven wheat genotypes were used to study the genetic diversity in response to drought stress. Under laboratory conditions, biochemical genetic markers, such as peroxidase (POX), ascorbate-peroxidase (APX) and malate dehydrogenase (MDH) were studied to determine the induction of drought responsive new isoforms, genetic diversity and polymorphism among wheat genotypes under control, 10% (−0.26 bar) and 20% (−0.521bar) polyethylene glycol (PEG-6000) stress conditions. Electrophoretic analysis of three isozymes revealed the specific appearances of some new drought responsive isozyme bands in wheat genotypes under stress conditions compared to the control. In some cases, observed banding pattern of isozymes were the same but density of bands was more under stress conditions than control in all the wheat genotypes. So, the results showed the activation of antioxidant defence mechanism upon stress condition. As a consequence, genotypes which showed favourable response to stress by inducing new isoforms could be indirectly considered as the promising biochemical genetic markers for the development of drought tolerant wheat genotypes.
