Infrared-Assisted Extraction of Adenosine from Radix Isatidis Using Orthogonal Experimental Design and LC

The present work reports a novel method for the extraction of adenosine from Isatis indigotica Fort. The extraction mechanism is based on rapid heating of extracting solvent and sample matrix under infrared radiation. Aqueous ethanol was used as extraction solvent. Extraction conditions of infrared-assisted extraction (IRAE) were optimized using orthogonal design, and the optimum conditions were as follows: infrared power of 260 W, radiation time of 45 min, extraction solvent of 40% aqueous ethanol and a solid/liquid ratio of 1:20 w/v. Under strong penetrability, significant heat effect and uniform heating, IRAE showed relatively higher yield of adenosine, when compared to conventional extraction techniques. A precise, accurate and sensitive method for the determination of adenosine in Radix Isatidis was also developed. The limit of quantification for adenosine was 0.12 μg mL⁻¹ with a regression coefficient of 0.9998. Extraction recoveries of adenosine were between 95 and 100%. These results suggested the potential of IRAE for the extraction of active compounds from a variety of matrices.
