Structure-function relationships in cellulase genes

Cellulases are mechanistically interesting because the hydrolysis of crystalline cellulose requires the action of a set of enzymes acting together, despite the fact that all of the enzymes catalyze the hydrolysis of a beta-1,4-linkage between two glucose residues. Genetic engineering techniques are being applied to cellulase genes in order to determine the mechanism of action of cellulases and to try to engineer more active cellulases. This review describes various approaches currently used to improve cellulases and defines new directions for recombinant cellulase technology.
