QTL mapping for disease resistance and fruit quality in grape

Two previous molecular linkage maps based on a population of Vitis vinifera Moscato bianco x Vitis riparia were supplemented with additional codominant markers and used to produce QTL information for resistance to downy mildew of the leaves and aroma compounds of the berries. For the maternal map, 403 SSR and AFLP markers were assembled in 21 linkage groups covering a length of 1128 cM while 502 SSRs and AFLPs were placed in 20 linkage groups of the paternal parent and 1143 cM of total map distance. Polymorphisms detected by the SSCP technique based on DNA sequence information of grape ESTs, and EST-derived microsatellite markers allowed to map 14 functional genes, some of them related to defense in plants. For downy mildew resistance trait, phenotypic data were evaluated in the segregating population after natural infections and artificial inoculations with Plasmopara viticola. Cuttings of the seedling population were also rated for sporulation, chlorosis and necrosis after artificial inoculation in greenhouse controlled conditions. Aroma composition of the berries was elucidated by HRGC-MS analysis for a subset of individuals grouped in neutral, slightly aromatic and aromatic by berry tasting. First detection of QTLs for downy mildew symptoms after artificial inoculation and berry content of free aroma forms is reported.
