Molecular breeding of a Brevibacterium flavum L-lysine producer using a cloned aspartokinase gene

The lysC gene encoding aspartokinase of a lysine-hyperproducing mutant Brevibacterium flavum CCRC 18271 was cloned by polymerase chain reaction and its expression in CCRC 18271 was studied. Enzyme assays showed that B. flavum cell harbouring the lysC gene exhibited 4- to 11-fold higher specific activities of aspartokinase as compared to respective hosts. Introduction of the lysC gene into B. flavum CCRC 18271 resulted in increased lysine production up to 33% in flask fermentations.
