Advances of melon hybrid purity test

Melon hybrid seed purity is the core of the seed industry. Quality is also the main basis and an important factor at seed level, which affects hybrid production and quality. The genetic purity test is an important process for melon hybrid seed production. This paper reviews methods of melon hybrid purity test with traditional methods for identifying field cultivation, isozyme electrophoresis of protein technology and DNA molecular marker technique. The traditional planting field survey method has the advantages of being intuitive, a technology which is easy to master, but it is vulnerable to environmental impact. The result is not accurate, the appraisal cycle is long and laborious for the judges. The planting field management requirements are also higher. The isozyme electrophoresis of protein technology method has a high sensitivity and thermal stability but it is not easy to interact with samples or easy to operate, however it has the application of fingerprint. With the development of molecular biology technology, molecular marker technology offers a new way for cultivar identification. DNA molecular markers include random amplified polymorphic DNA (RAPD) technology, amplified fragment length polymorphism (AFLP) technology, simple sequence repeats (SSR) technology, sequence-related amplified polymorphism (SRAP) technology, etc. These DNA molecular markers could shorten the cycle of identification and the results are more accurate. For melon hybrid seed purity identification and fingerprint application, SSR molecular markers are gaining more and more attention as a method to be used, because they have the characteristics of high resolution and codominance.
