An enzyme-linked immunosorbent assay has been developed for the detection of low levels of methoprene from tobacco samples. The generation of anti-methoprene antibodies needed for such an assay relied on the preparation of a methoprene-carrier immunogen. Methoprene acid was covalently bound via an ester to a spacer group, which in turn was bound to a protein carrier. Two activated ester methods were used to prepare the immunogen, one of which forms a water soluble, activated ester of methoprene. The resulting polyclonal antibodies raised against the methoprene immunogen were highly specific for methoprene, and did not cross react with closely related esters such as the insect juvenile hormones. The range of the methoprene ELISA was from 5 to 300 ng/mL, with an I50 of 50 ng/mL.
